TG101209
TG101209 is a novel and potent JAK2 inhibitor that induces dose- and time-dependent cytotoxicity in multiple multiple myeloma (MM) cell lines. Exploration of the mechanism of action of TG101209 revealed that pJak2, pStat3, and Bcl-xl levels were downregulated, while pErk and pAkt levels were upregulated, indicating the existence of crosstalk between signaling pathways. TG101209 demonstrated synergistic cytotoxicity against myeloma cells when combined with the PI3K inhibitor LY294002.
