Capecitabine is a pre-prodrug of 5-fluorouracil (5-FU). After oral uptake, capecitabine is first metabolized to 5'-deoxy-5-fluorocytidine (5'-DFCR), which happens mainly in the liver by carboxyl-esterase. The metabolite is converted to 5'-deoxy-5-fluoro-uridine (5'-DFUR) by cytidine deaminase in liver and tumor tissue. Then 5'-DFUR is converted to 5-FU intracellularly by thymidine phosphorylase, which is often expressed in tumor tissue. Capecitabine inhibits tumor growth by incorporation of 5-FU into RNA and DNA and inhibition of thymidylate synthase. Capecitabine is the active ingredient in the drug sold under the trade name Xeloda®. This drug has been approved in at least one country for use in patients with metastatic breast and colorectal cancers. NOTE: The capecitabine product sold by LC Laboratories is NOT Xeloda®, and is NOT for human use. Other CAS numbers previously assigned to capecitabine, 158798-73-3 and 958887-39-3, have been deleted by CAS and are no longer in use.
