Anti-CD33 immunotoxin (scFv)-Gel

This immunotoxin was achieved by conjugating/fusing the Anti-CD33 scFv to gelonin. It was expressed in CHO and purified with affinity chromatography. The immunotoxin selectively binds to acute myeloid leukaemia cells. Once bind, the toxin enters cells by receptor-mediated endocytosis. Then, gelonin, like abrin and ricin, inhibits protein synthesis by damaging the 60S subunit of mammalian ribosomes. It was designed for treating acute myeloid leukaemia.
