Birinapant, also known as TL32711, is an antagonist for the inhibitor of apoptosis protein (IAP). IAP promotes cancer cell survival and confers resistance to cancer therapy. Birinapant induced apoptosis and enhanced TRAIL potency in inflammatory breast cancer cells. Birinapant in combination with TNF-α exhibited a strong anti-tumor effect in 12 of 18 melanoma cell lines and was effective in a melanoma cell line with acquired resistance to BRAF inhibitors. Birinapant, a bivalent SMAC mimetic, targeted TRAF2-associated cIAP1 and cIAP2, blocked TNF-induced NF-κB activation, and was active in multiple primary patient-derived xenograft models.
