Mouse Anti-MIC Recombinant Antibody (clone 1D5); scFv Fragment
The 1D5 antibody shows a complete loss in binding to the MICA*008 a3 domain containing either the glycosylation variant Glycol 4 (G243N) or Glycol 6 (R279N), but is unaffected by Glycol 1 (R202N), Glycol2 (E215N), Glycol3 (I236T), Glycol5 (H248N), and Glycol7 (C-terminal addition of residues N298, G299, S300). Comparison of the epitope of 1D5 determined by X-ray crystallography with the location of the glycosylation site variants demonstrates that Glycol 4 (G243N) and Glycol6 (R279N) are part of the 1D5 epitope, whereas Glycol 1 (R202N), Glycol2 (E215N), Glycol3 (I236T), Glycol5 (H248N), and Glycol7 (carboxy-terminal addition of residues N298, G299, S300) fall outside of the 1D5 epitope. Binding of 1D5 or 13A9 to the MICA a3 domain may create steric hindrance and block access of proteases required for cleavage of MICA.
